In full:
The "Α" in "άμεμπτος" is an example of "A privative".

Privative i.e. without whatever the rest of the word indicates - in this case blame. So, "without blame, blameless".

It is important to distinguish this from e.g. the A in the nickname Αχειλάς (euphonic, perhaps), as Αχειλάς certainly does not mean "without lips".
