Using a sterile loop, spread the aliquot of liquid over the quadrant.
Using sterile loops, perform a three-zone streak to isolate any bacteria (Figure 1).
Incubate the agar plates at 36 ± 2°C for 24 hours.
Inspect each plate for colonial morphology and purity.
Should mixed cultures be suspected (varied colonial morphology) additional single colony subcultures should be considered.
NOTE:
If different colony types are well isolated, secondary confirmation may be performed without additional subculture.
